This preprint reports laboratory work showing that sustained fluid shear stress can trigger normal human intestinal fibroblasts to switch into a profibrotic, mechanoadaptive state in engineered shear platforms (a microfluidic gut-on-a-chip and a mesofluidic rotary shaker).

The switched cells formed 3D aggregates with strong α–smooth muscle actin expression and extracellular-matrix features seen in fibrotic tissue; proteomics showed increases in collagens and matrix metalloproteinases. The experiments were done in controlled in vitro platforms and focus on cellular and ultrastructural readouts (electron microscopy, proteomics).
