In cultured human intestinal epithelial cells BPA impaired mitochondrial function (lower membrane potential, Complex I activity, ATP; higher mitochondrial superoxide) and increased cleaved CASP1, GSDMD-N, IL‑1β and NF-κB signaling.
The authors used a CASP1 inhibitor (VX-765) and computational analyses: pharmacologic CASP1 blockade partially preserved proliferation and barrier proteins but did not fully reverse inflammatory or metabolic stress effects.
Computational docking and human single-cell transcriptomic reanalysis supported CASP1 as a plausible BPA-associated target and showed CASP1-high epithelial cells in IBD enriched for inflammatory/pyroptosis programs.
