What To Know

This study (abstract) reports that a pectic polysaccharide extracted from rhubarb (RP-2) reduced inflammation and disease severity in a DSS-induced mouse model of ulcerative colitis. The authors characterized RP-2 structurally, observed prolonged retention in inflamed colon tissue, and showed microbiota changes (enrichment of SCFA-producing bacteria).

Antibiotic depletion reduced RP-2’s protective effects, and mechanistic experiments implicate suppression of the TLR4/MyD88/NF-κB pathway and restoration of Th17/Treg balance. RP-2 effects were tested in cell culture (RAW264.7 macrophages) and in mice with chemically induced colitis; the abstract does not report human data.

The protective effect depended on the gut microbiota and was associated with improved barrier markers and reduced pro-inflammatory cytokines in the model. This is a preclinical, mechanistic report of a natural product in animals and cells; it suggests a microbiota–metabolite–immune–barrier axis but does not provide clinical efficacy or safety data in people.
