What To Know

The paper used network toxicology, molecular docking, and mouse experiments (DSS colitis model) to identify three protein targets (MMP9, PTGS2, PPARG) that bind 6PPDQ and change in expression after exposure.

On its own, 6PPDQ did not visibly damage mouse colon, but mice pre-exposed to 6PPDQ developed more severe DSS-induced colitis by several measures (weight loss, higher disease activity index, colon shortening, higher MPO and pro-inflammatory cytokines, lower IL-10).

The study is a preclinical toxicology and mechanistic investigation in mice and in silico analyses. It identifies plausible molecular targets and demonstrates an interaction between a common environmental contaminant and an experimental colitis trigger, but it does not demonstrate direct effects in humans.
