The authors compared wild-type and Adamdec1−/− mice co-housed in two different environments and induced colitis with DSS. Severe disease in Adamdec1−/− mice was linked with greater neutrophil recruitment, altered myeloid activation (CD11b, monocyte/macrophage changes), impaired epithelial proliferation, and disrupted stem cell–associated gene expression.
Distinct fecal microbiome profiles tracked with these phenotypic differences, supporting a gene–environment–microbiome interaction. The study suggests that in genetically susceptible hosts, environmental and microbial factors can determine colitis severity, and that modifying the microbiome might be a strategy worth exploring.
